Mesenchymal stromal cells in the bone marrow niche consist of multi-populations with distinct transcriptional and epigenetic properties

Abstract Although multiple studies have investigated the mesenchymal stem and progenitor cells (MSCs) that give rise to mature bone marrow, high heterogeneity in their morphologies properties causes difficulties molecular separation of distinct populations. In this study, by taking advantage resolution single cell transcriptome, we analyzed Sca-1 PDGFR-? fraction mouse marrow tissue. The transcriptome enabled us further classify population into seven populations according gene expression profiles. We then separately obtained based on candidate marker genes, specified epigenetic landscape ATAC-seq. Our findings will enable elucidate niche signal microenvironment, reconstitute vitro, shed light potentially important role identified subpopulation various clinical applications treatment bone- marrow-related diseases.
